Vincent D'Onofrio was born on June 30, 1959, in Brooklyn, New York. Growing up, his family frequently moved from place to place, which allowed Vincent to experience different environments such as Hawaii, Florida, and Colorado. In 1977, he graduated from Hialeah-Miami Lakes High School in Florida and later enrolled at the University of Colorado. However, he dropped out after 18 months and moved to New York to pursue a career in acting.

In New York, D'Onofrio joined the American Stanislavsky Theatre and became a full-fledged actor in 1984. He performed in various plays such as "Of Mice and Men" and "Sexual Perversity" and made his Broadway debut in "Open Admissions" as Nick Rizzoli. His natural talent and extensive theater experience contributed to his success on stage.
