Antonio Rüdiger's journey into professional football began in 2011 when he joined the academy of German club VfB Stuttgart. He made his debut for the club's reserve team, Stuttgart II, in the Third Bundesliga in July 2011.

In January 2012, Rüdiger earned his first call-up to the Stuttgart senior team and made his debut in a Bundesliga match against Borussia Mönchengladbach. He continued to impress and made his debut in the Europa League in October 2012. Rüdiger extended his contract with Stuttgart until 2017 in April 2013.
