Jílek was born in Brno, Czech Republic, in 1920. He first studied music under Zdeněk Chalabala in Brno before continuing his education at the Prague Conservatory with Vítězslav Novák.

Jílek's career in opera began in 1936 when he joined the Brno Opera as an assistant. In 1939, he became the chief conductor of the Ostrava Opera. After World War II, he returned to Brno in 1948, initially as assistant conductor and later as chief conductor from 1952 to 1978.
