Călinescu pursued Italian studies at the University of Bucharest and later taught there as a professor. He also held the position of director of the Institute of Literary History and Folklore, an institution that now bears his name. In 1946, he became an academician of the Romanian Academy.

Călinescu's literary works include seminal studies on Romanian authors such as Mihai Eminescu, Ion Creangă, and Nicolae Filimon. His five-volume biography of Eminescu, entitled "The Life of Mihai Eminescu" (1934-1936), is considered a masterpiece of literary scholarship.
