In her childhood, Madhuri did not dream of becoming an actress or a dancer, but she pursued Kathak dancing. She attended school and later enrolled in the Parle College in Mumbai to study microbiology. After obtaining her bachelor's degree, Madhuri finally considered a career in acting. She is one of the few Bollywood actresses of the 80s who received a complete school education and has a bachelor's degree in microbiology.

In 1986, she accepted an offer from Rajshri Productions to star in the film "Abodh." The film did not perform well, and Madhuri went unnoticed, leading her to take small roles and dance numbers. One of these roles was given to her by director Subhash Ghai in the film "Karma" (1986), where she received critical acclaim. Ghai even placed advertisements featuring Madhuri in several magazines, claiming that prominent filmmakers like Boney Kapoor and Yash Chopra had signed contracts with her, although it was not true. Madhuri also appeared in Ghai's film "Ram Lakhan" (1988), which was expected to be her first hit. However, it was the energetic dance number "Ek, do, teen" in N. Chandra's "Tezaab" that catapulted her career. "Tezaab," not "Ram Lakhan," turned her into a sensation, and the Bollywood industry took notice of her talent.

The 90s became the most successful period of Madhuri's career. In 1990, she starred in "Dil," which earned her the first Filmfare Award for Best Actress. Her on-screen pairing with actor Anil Kapoor proved successful in films like "Beta" (1991), which brought her a second Filmfare Award, and "Raja" (1995). Following "Raja," she earned the nickname "The Amitabh Bachchan of Women" after her performance in the film "Koyla." In 1994, her film "Hum Aapke Hain Koun" became the biggest blockbuster in Indian cinema, and she won another Filmfare Award for her role.

However, in 1995, Madhuri's success began to decline with a series of unsuccessful films such as "Yaarana," "Rajkumar," "Prem Granth," and "Mahaanta," despite receiving critical praise for her performances. Director Rakesh Roshan even made negative comments about her ability to play young heroines after the film "Koyla." In 1997, she regained her previous fame with the films "Mrityudand" and "Dil To Pagal Hai," which became the highest-grossing film of 1997, earning her another Filmfare Award.

In the late 90s, Madhuri's films like "Wajood," "Gajagamini," "Pukar," and "Engineer" were well-received by the audience and solidified her status as a superstar. In 2002, she accepted director Sanjay Leela Bhansali's invitation to play the role of Chandramukhi, a courtesan, in the film "Devdas," which was showcased at the Cannes Film Festival and became a landmark in Indian cinema.

In 2007, Madhuri starred in the dance drama film "Aaja Nachle" produced by Aditya Chopra's Yash Raj Films. The film was moderately successful at the box office. In June 1999, Madhuri met Shriram Nene, a cardiovascular surgeon of high caste from Maharashtra who had spent most of his life in the United States. They got married on October 17 in Laguna Beach, away from the media's attention. Madhuri relocated to Denver, USA, and embraced a life as a homemaker.

Madhuri became a mother in March 2003 when she gave birth to her first son, Arin. She prioritized her child and decided to settle in America, leading a normal domestic life. In March 2005, she gave birth to her second son, Ryan.
