Before entering the House of Representatives in 1991, DeLauro served as an administrative assistant, chief of staff for Senator Chris Dodd, and executive director of the EMILY's List committee. In 1990, Congressman Bruce Morrison's decision to run for governor of Connecticut created a vacancy in the lower house of Congress. DeLauro won the seat by defeating Thomas Scott with a 52% to 48% margin. She never faced such a close margin again and was re-elected to the House ten times, always receiving at least 63% of the votes.

DeLauro is known for her liberal stance and is a founding member of the Congressional Progressive Caucus. She has been actively involved in healthcare issues, particularly women's health. DeLauro has introduced numerous bills aimed at advancing medical research and improving cancer treatment. She harshly criticized the Food and Drug Administration when it refused to implement a program to protect the population from unsafe food and drugs.

On February 2, 2008, DeLauro endorsed Senator Barack Obama as a candidate for president. On October 3, 2008, she voted for the Emergency Economic Stabilization Act of 2008, which aimed to help overcome the economic crisis. DeLauro has also called on politicians to be "more serious thinkers" when it comes to universal healthcare issues. In December 2008, The Wall Street Journal named DeLauro as a top contender for the position of Secretary of Labor in the Obama administration. However, President Obama appointed congresswoman Hilda Solis to the position on December 19.

DeLauro has been a vocal critic of the Stupak-Pitts amendment, which sought to significantly change abortion rights in the United States. She opposes the prohibition of abortions in cases of incest, rape, or when the life of the pregnant woman is at risk. Additionally, she supports the use of federal subsidies for abortions. In 2006, she voted against the HR 6099 bill, which required certain procedures and formalities to obtain permission for an abortion. These positions have drawn criticism from the Roman Catholic Church, of which DeLauro is a member.
