He gained recognition for his portrayal of Sebastian in the classic science fiction film "Blade Runner" directed by Ridley Scott. Additionally, Sanderson captivated audiences with his character Deuce in the science fiction television series "Babylon 5" and its sequel "Babylon 5: Thirdspace".

Sanderson's career took off in the early 1980s when he played Larry, a recurring character in the American sitcom "Newhart" from 1982 to 1990. His signature line, "Hi, I'm Larry. This is my brother Darryl, and this is my other brother Darryl," became iconic in the show. He regained prominence on the small screen with his role as E.B. Farnum, the comedic yet tragic hotel owner and first mayor of Deadwood, in the series "Deadwood" from 2004 to 2006.
